Abstract


Medicinal plants are widely used in Africa, especially in rural areas, to meet primary health care needs. Among them Massularia acuminata (Rubiaceae) is mainly used for its aphrodisiac properties.In order to verify its acute toxicity, a single dose of 5000 mg/kg body weight (bw) of the ethanolic extract of the stems of this plant was administered to a group of 10 female rats. The control group, also consisting of 10 female rats, received only distilled water. During the 14 days observation, all clinical signs and deaths were reported. At the end of the study, the LD50, blood hematological and biochemical parameters, and hepatic and renal histological examination were performed.The results showed that the administration of the ethanolic extract of the stems of Massularia acuminata did not modify the behavior of the rats and the estimated LD50 was higher than 5000 mg/kg bw. The evaluation of hematological and biochemical parameters revealed a significant decrease (p <0.05) in the level of white blood cells and platelets, and a significant increase (p <0.05) in creatinine and total cholesterol. As for the histological study, apart from some hepatic apoptosis, the result did not show any hepatic and renal lesions.This study showed that the ethanolic extract of the stems of Massularia acuminata could exhibit hematotoxicity, nephrotoxicity and hepatotoxicity when higher doses were used.

Les plantes médicinales sont largement employées en Afrique notamment dans les milieux ruraux pour satisfaire les besoins de soins primaires. Parmi les plantes utilisées, figure Massularia acuminata (Rubiaceae) recherchée principalement pour ses vertus aphrodisiaques.Afin de vérifier sa toxicité aiguë, l’extrait éthanolique des tiges de cette plante a été administré à une dose unique de 5000 mg/kg de poids corporel (pc) à un groupe de 10 rats femelles. Le groupe témoin constitué aussi de 10 rats femelles n’a reçu que de l’eau distillée. Durant les 14 jours d’observation, tous les signes cliniques et les décès ont été notifiés. A la fin de l’étude, la DL50, des paramètres hématologiques et biochimiques sanguins et l’examen histologique hépatique et rénal ont été réalisés.Les résultats ont montré que l’administration de l’extrait éthanolique des tiges de Massularia acuminata ne modifie pas le comportement des rats et la DL50 estimée a été supérieure à 5000 mg/kg de pc. L‘évaluation des paramètres hématologiques et biochimiques ont mis en évidence une baisse significative (p < 0,05) du taux de globules blancs et des plaquettes sanguines, et une augmentation significative (p < 0,05) de la créatinine et du cholestérol total. L’étude histologique, hormis une apoptose hépatique, n’a provoqué de lésions hépatique et rénale.Cette étude a permis de montrer que l’extrait éthanolique des tiges de Massularia acuminata pourrait à forte dose présenter une hématotoxicité, une néphrotoxicité et une hépatotoxicité.
