Abstract


The objective of this study is the production of friable embryogenic callus in citrus to having the necessary material for protoplast isolation to be integrated into rootstock improvment programm using the asexual pathway. Ovules, embryos and styles explants of Citrus five genotypes: Poncirus trifoliate, Citrus reshni Hort. ex Tan, Citrus amblicarpa, Citrus volkameriana and Citrus paradisi Macf., are cultured in MT medium supplemented with 4% sucrose, 0.05% of malt extract and 1% agar was used as basal medium. Hormones (kinetin (Kn), 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP) were added to this medium at different concentrations. The rate of callus of different explants was evaluated by determining the percentage of callus induction by explant. The conducted experiments show a significant effect of genotype, explant type and hormone combination chosen. Friable callus forms were on the 3 explants with variable frequencies. Embryo explants show is most favorable to the production of friable callus; the best callogenesis is obtained with the culture media containing auxin and cytokinin combined M2 (1mg/l 2,4D+ 0.5 mg/l BAP) and M3 (1mg/l2,4D + 0.5mg/l Kn), by against the style explants and ovule have induced embryogenic calli respectively on the M4 (0.5 mg/l BAP) and M5 (1mg/l Kn) medium.

L’objectif de cette étude est la production de cals friables embryogènes de différents génotypes d’agrumes afin de disposer d’un matériel de base destiné à être intégré dans un programme d’amélioration de porte-greffes par voie asexuée. Des explants d’ovules, d’embryons et de styles de cinq génotypes d’agrumes : Poncirus trifoliata, Citrus reshni Hort. ex Tan, Citrus amblicarpa, Citrus volkameriana, et Citrus paradisi Macf., ont été mis en culture sur le milieu MT additionné de 4 % de saccharose, de 0,05 % d’extrait de malt et de 1 % d’agar, a été utilisé comme milieu de base. Les hormones (kinétine (Kn), acide 2,4-dichlorophénoxyacétique (2,4-D) et 6-benzylaminopurine (BAP) ont été ajoutées à ce milieu à différentes concentrations. Le taux de callogenèse de différents explants est évalué par la détermination du pourcentage d’induction des cals par explants. Les expérimentations mettent en évidence un effet marqué du génotype, du type d’explant et de la combinaison hormonale choisie. Des cals friables ont été produits sur les 3 explants avec des fréquences variables. Les explants d’embryon semblent plus favorables à la production des cals friables ; la meilleure callogenèse a été obtenue avec les milieux de culture additionnés d’auxine et de cytokinine combinés M2 (1mg/l 2,4-D+ 0.5 mg/l BAP) et M3 (1mg/l2,4-D + 0.5mg/l Kn). Par contre les explants de style et d’ovule ont induit des cals embryogènes respectivement sur le milieu M4 (0.5 mg/l BAP) et M5(1mg/l Kn).
